CAIX (Carbonic anhydrase IX) is a hypoxia and HIF-1–inducible protein that regulates intra- and extracellular pH under hypoxic conditions and promotes tumor cell survival and invasion in hypoxic microenvironments. U-104 is a novel ureido-sulfonamide inhibitor of CAIX and CAXII with Ki values of 45.1nM and 4.5nM, respectively. Administration of U-104 daily significantly inhibited primary tumor growth in the mice injected with a highly metastatic variant of the MDA-MB-231 cell line with robustly induced CAIX in hypoxia. Also, it resulted the inhibition of metastases formation of mice intravenous injected with 4T1 cells.
