Angiotensin Receptor (ATR) is a kind of G protein-coupled receptor (GPCR) and plays an impotent role in multiple cellular functions, such as blood pressure, thirst, sodium balance, growth-promoting, proinflammatory, profibrotic actions and so on. Angiotensin II, as a the octapeptide hormone, could at least interact with two high-affinity receptors, designated as AT1R and AT2R. Consistent with interaction with AT1R by angiotensin II, with the addition of 1*10-7M angiotensin II, the level of vascular endothelial growth factor (VEGF) mRNA in rat heart-derived endothelial cells would increase 5-fold at 2h and then decline over 4 h, which was mainly attributed to the transcriptional activation by enhancing AP-1 and NF- κB binding. Meanwhile, correlating well with its mRNA level, the intensity of some certain VEGFs was increased from 8 to 12h with treatment of angiotensin II in the conditioned medium, which had been shown to promote the growth of endothelial cell and change its permeability. Continuous infusion of 1ug/kg/min angiotensin II for 14 days could induce a marked increase of VEGF in distal tubules and a moderate increase in proximal tubules of male Sprague-Dawley (SD) rats, as well as elevate the level of flk-1, a major VEGF receptor, for 1.8-fold compared to the PBS- or NE-infused group through AT1R. Besides of the interaction with AT1R, continuous infusion of angiotensin II could also result in interstitial fibrosis in the rats through AT2R, which could be slightly diminished by hydralazine, a AT2R receptor antagonist.
