MMAE is a potent synthetic antimitotic agent working as microtubule inhibitor. It is usually linked to specific monoclonal antibodies like the chimeric mAbs cBR96 (specific to Lewis Y on carcinomas) and cAC10 (specific to CD30 on hematological malignancies), used as a payload of ADCs. MMAE exhibited potency against cell growth with GI50 values of 0.22 nM, 0.49 nM and 0.54 nM for BT474, MDA-MB-361-DYT2 and N87 cells, respectively. Through combination with specific monoclonal antibody through specific linker, like antibody-drug conjugate cAC10-vcMMAE, the cytotoxicity and selectivity of MMAE was significantly enhanced. Intravenous injection with cAC10-vcMMAE at dose of 1mg/kg induced complete, durable tumor regression in the Karpas 299 ALCL model.
