Topotecan, a water-soluble semisynthetic analogue of the alkaloid camptothecin, is a potent Topoisomerase I inhibitor. Topotecan could stabilize topoisomerase I-DNA cleavable complexes in radiation-resistant human B-lineage acute lymphoblastic leukemia cells. Exposure to 50 or 100nM Topotecan for 5 days induced apoptotic cell death of radiation-resistant RS4;ll leukemia cells despite of the high levels of bcl-2 protein, without change in bcl-2 protein levels. Infusion with 200μg or 400μg Topotecan could enhance the survival ratio of SClD mice inoculated with RS4;ll. LC1:19, or NALM-6 leukemia cells. Topotecan showed more potency against neuroblastoma cell viability with amplification of MYCN, due to the disruption of B-MYB-MYCN axis and selective down-regulation of B-Myb and MycN expression by Topotecan in IMR-32, Kelly, LAN-5 and SK-N-BE 2 cells.
